Does anyone know of a mapping app that will let you plot a cycle route (on and off road) and then work as a normal sat nav guiding you around the route you have plotted?

Have a look at Viewranger.

Both strava and Endomondo will do it, best one i found was the Garmin one.

Viewranger charges for the maps. Strava I can plot my route but I need an android app that will then play the route back to me aka sat nav style as I cycle. Any suggestions?

You'll need a decent Garmin or one of the paid Apps like OS maps

The trailforks app lets you draw a route (not sure if it can do it by gps, think it's by hand), and then should supposedly show your real time location on the map, but I've never found a signal is good enough to make it smooth, so it's not brilliant.

You don't have to buy maps for Viewranger, you can create routes on open cycle maps or viewranger landscape maps and download them to your phone. They are both worldwide maps, I've used them on the fly in France a few times but for that you need a good signal. It's better to download them beforehand.
